The SB-100 mRNA expresses the Sleeping Beauty transposase (sequence reference: Jin Z, et al. Gene Therapy, 18, 849-856 (2011)). Sleeping beauty transposon and transposase constitute a DNA plasmid system used for therapeutic human cell genetic engineering. This mRNA is capped with Cap1 structure with high capping efficiency. It has 100% substituted with N1-methyl-pseudoUridine for enhanced expression and reduced immunogenicity. The mRNA has a 100A tail in its sequence, mimics a mature mRNA.

Description

The SB-100 (Sleeping Beauty) mRNA expresses the Sleeping Beauty transposase. Sleeping beauty transposon and transposase constitute a DNA plasmid system used for therapeutic human cell genetic engineering. The SB-100 mRNA is hyper sensitive, sequence reference from publication Jin Z, Maiti S, Huls H, et al. Gene Therapy, 18, 849-856 (2011).  
 
This mRNA features: 
- A Cap 1 structure with high capping efficiency 
- 100% substitution with N1-methyl-pseudouridine (m1Ψ) for improved protein expression and reduced innate immune response 
- GenScript’s patented UTR to enhance translation 
- A 100A poly(A) tail to mimic natural mature mRNA

Applications
Seed 5 × 10⁴ HEK293T cells per well in a 24-well plate one day before transfection. On the day of transfection, replace the culture medium with OptiMEM supplemented with 10% FBS. Prepare a transfection mix containing SB100 mRNA and donor DNA (recommended molar ratio 1:2, total amount 1μg), and deliver using Lipofectamine 2000 (or equivalent). After transfection, passage cells on Day 3, 6, 10, and 12. Collect samples at each time point for flow cytometry analysis.
