Cells were incubated with Calcium-4 and indicated concentrations of capsazepine for 1 hour, then stimulated with 50 μM capsaicin. Calcium responses were recorded on a FlexStationIII plate reader. Cells were loaded with membrane potential sensitive dye (Molecular Devices) and then stimulated with the indicated concentrations of ATP. (B) Dose-response relationship of TN-ATP to 293/P2X3 cells. Cells were incubated with membrane potential sensitive dye (Molecular Devices) and indicated concentrations of TN-ATP for 45 minutes, then stimulated with 50 μM ATP. Calcium responses were recorded on a FlexStationIII plate reader. Data represent the average +/- standard deviation of triplicate determinations.
